The project starts from the consideration that many patients of idiopathic chronic pancreatitis have at least one CFTR mutation and intends to analyze the role of the CFTR protein in the determinism of the disease, with particular reference to the transport of bicarbonate. The study will use clinical tests used in the diagnosis of atypical CF to evaluate the CFTR function, such as the test of nasal potentials, the intestinal short circuit current, the sweat flow test (bubble test). But above will be used the intestinal organoids (microscopic structures produced from stem cells of epithelium obtained from rectal biopsy). The swelling of the organoids in response to stimuli of the CFTR channel will give the measure of how much CFTR mutated is involved in the genesis of pancreatitis. Particular attention will be paid to the measurement of the bicarbonate flow, notoriously compromised in CFTR defects. Other techniques, such as confocal reflectometry and the FIS technique will complete the study on organoids. The ultimate aim of the project is to develop possible new treatments of idiopathic chronic pancreatitis.
