Given the well-established role of phosphoinositide 3-kinase γ (PI3Kγ) as negative regulator of cAMP (Project FFC#25/2014) this new project intended to explore the therapeutic potential of PI3Kγ inhibition in cell-based and pre-clinical models of CF. It was planned to evaluate whether a cell-permeable peptide targeting PI3Kγ activity (Patent pending N° TO2014A001105) corrects the primary cause of CF by potentiating the activity of the mutant channel in epithelia. In addition, it aimed to explore whether the same compound was effective in inhibiting leukocyte activation, thus reducing the uncontrolled lung inflammation that typically occurs in CF patients. Finally, the researchers intended to evaluate the ability of the molecule to serve as bronchodilator, thus alleviating CF-associated bronchial hyperresponsiveness. Inhibition of PI3Kγ scaffold function via aerosolized formulation of Trojan peptides might thus provide a novel therapeutic strategy.
